I. Background
由 IL-12、IL-23、IL-27、IL-35 和 IL-39 形成的细胞因子白细胞介素 12 （IL-12）家族在先天性和适应性免疫应答的诱导和调节中起关键作用。IL-27 主要由抗原呈递细胞（APC）产生，包括树突状细胞（DC）、浆细胞、巨噬细胞、炎性单核细胞、小胶质细胞、B 细胞和内皮细胞。IL-27 通常参与不同 CD4 T 细胞亚群的分化和激活。受肿瘤抗原和 CD40 刺激的抗原呈递细胞可以分泌 IL-27，IL-27 在调节针对癌症的免疫反应方面具有双重功能（促炎和抗炎作用）。IL-27 对人类病毒具有抗病毒特性，包括流感、单纯疱疹病毒、人类乙型肝炎（HBV）和丙型肝炎（HCV）、人类免疫缺陷病毒（HIV）和其他病毒等。 IL-27 是一种由 EBi3 和 p28 亚基组成的异二聚体细胞因子。IL-27 通过由两个亚基组成的异二聚体受体发出信号，即 IL-27Rα（也称为 WSX-1 或 TCCR）和 gp130 （IL6ST）。 IL-27 结合激活 Janus 激酶（JAK）信号转导和转录激活因子（STAT1 和 STAT3）和丝裂原活化蛋白激酶（MAPK）信号转导通路。
